The study identifies and characterizes a novel multidrug resistance operon, mdtRP (yusOP), in Bacillus subtilis. The mdtR gene encodes a MarR-type transcriptional regulator, and the mdtP gene encodes a multidrug efflux transporter. Mutations in mdtR, such as Arg83 → Lys and Ala67 → Thr, lead to multidrug resistance by derepressing the mdtRP operon.
